L-2286
L-2286 is an orally active PARP-1 inhibitor. L-2286 alleviates carotid artery remodeling, oxidative stress and inflammation in spontaneously hypertensive rats, protects neurons in the dorsal hippocampus, and reduces pyramidal cell loss and gliosis without affecting blood pressure. L-2286 can be used in research related to hypertension.

Animal Model:Spontaneously Hypertensive Rats (SHR) (10-week-old male, hypertension model)[1]
-
Dosage:5 mg/kg/day
-
Administration:administered via drinking water for 32 weeks
-
Result:Significantly reduced carotid intima‑media thickening, interstitial fibrosis, oxidative/nitrosative stress, nuclear factor‑κB (NF‑κB) nuclear translocation, and apoptosis‑inducing factor (AIF)‑dependent cell death in carotid arteries, while improving endothelium‑dependent vasodilation in spontaneously hypertensive rats.
Mitigated oxidative damage, pyramidal neuron loss, DNA oxidation, poly(ADP‑ribose) polymerase (PAR) formation, reactive astrogliosis, and perivascular white matter lesions in the dorsal hippocampus, all without altering systolic blood pressure.
