TZ3O is an anticholinergic agent with neuroprotective effects. TZ3O inhibits acetylcholinesterase (AChE) activity in human plasma with an IC50 of 304.5 μM. TZ3O can improve memory impairment and cognitive decline in rats in the Scopolamine (HY-N0296)-induced Alzheimer-type model. TZ3O could be used in Alzheimer’s research.

Animal Model:Scopolamine (3 mg/kg, i.p.) was administered to adult male Wistar rats (250-300 g) to induce cholinergic neurotransmission disorder, constructing an Alzheimer’s disease model with memory and cognitive impairment[1]

Dosage:0.5, 1, 2, 4 mg/kg

Administration:i.p., 60 min before trials, 8 days total with 2 rest days

Result:Reduced escape latency in Morris Water Maze test.
Increased spontaneous alternation behavior in Y-maze task.
Increased Nissl-stained neurons in hippocampal CA1/DG and cortex.
Upregulated mAChR M1 expression in hippocampal CA1/DG and cortex.
Reduced TUNEL-positive cells in hippocampus and cortex to inhibit neuron apoptosis.
Lowered AChE activity and relative expression in hippocampus and cortex.
Suppressed caspase 3 and caspase 8 activity in hippocampus and cortex.
Decreased plasma IL-6 and TNF-α levels.
Decreased hippocampal p-Tau and HSP70 levels.
Increased hippocampal BDNF, Nrf2 expression and hippocampal mAChR M1 protein expression.
Reduced plasma AST, ALT, and ALP levels.
