MV14
MV14 is a small-molecule NO donor that exhibits selective antiproliferative activity against bladder cancer cells. MV14 releases NO, causing DNA double-strand damage, inhibiting the NF‑κB pathway, and downregulating the apoptosis-inhibitory protein Survivin. MV14 interferes with the cell cycle and promotes tumor cell apoptosis. MV14 attenuates tumor-associated histopathological changes, urothelial destruction, and stromal invasion in the BBN (HY-W755252)-induced mouse bladder cancer model, and reduces PCNA levels in bladder tumor tissue while increasing active caspase-3. MV14 can be used for research on bladder cancer.

In Vitro
MV14 (0.19-6.25 μM; 24 h) exerts antiproliferative activity against T24 and 253J bladder cancer cells that is partly contributed by NO release, with T24 cells showing higher sensitivity to NO scavenging[1].
MV14 (0.39-3.13 μM; 24 h) inhibits the migration of T24 bladder cancer cells[1].
MV14 (5-10 μM; 3 h) induces concentration-dependent γ-H2AX DNA damage in T24 and 253J bladder cancer cells, and NO release is involved in this effect[1].
MV14 (0.39-1.56 μM (T24); 0.09-0.39 μM (253J); 24 h) reduces nuclear survivin in T24 and 253J bladder cancer cells[1].
MV14 (1.56-3.13 μM; 24 h) reduces total survivin protein in T24 bladder cancer cells[1].

-
Animal Model:C57BL/6 (female, 6-8 weeks old, received 0.05% BBN (HY-W755252) in drinking water ad libitum for 16 weeks)[1]
-
Dosage:1 mg/mL; 100 μL per intravesical instillation
-
Administration:intravesical; once weekly; four consecutive weeks (weeks 17-21)
-
Result:Attenuated tumor-associated histopathological alterations in BBN-induced bladder tumors, with less urothelial disruption and less pronounced stromal invasion than vehicle-treated tumor-bearing mice.
Tumors showed urothelial cell hyperplasia with lower-grade undifferentiation and dysplasia toward the lamina propria, moderate anisocytosis and anisokaryosis, nuclear atypia, fewer binucleated cells, and fewer mitotic figures than vehicle-treated tumors.
Reduced survivin levels and decreased PCNA-positive cells in tumor tissue, while increasing active caspase-3-positive cells.
