SM-17 is a human monoclonal antibody targeting IL-17RB. SM-17 binds to human IL-17RB, inhibits the dimerization of IL-17RA/IL-17RB, blocks IL-25-induced phosphorylation of ERK1/2 and STAT5, and suppresses Th2-mediated inflammatory responses. SM-17 inhibits the secretion of IL-5, IL-9, IL-13 and IL-4, impairs the proliferation/expansion of ILC2 and Th2 cells, and restores filaggrin expression. SM-17 reduces immune cell infiltration, epidermal hyperplasia, goblet cell hyperplasia, collagen deposition, ear swelling and scratching behavior in animal models. SM-17 can be used in research related to atopic dermatitis and allergic asthma.

SM-17 (5 μg/mL; 3-7 days) suppresses IL-5, IL-9, and IL-13 secretion from stimulated human PBMCs, isolated T helper cells, and enriched ILC2s after 3 or 7 days of incubation[1].
SM-17 (5 μg/mL) inhibits the proliferation of alarmin cocktail-stimulated Th2 cells and ILC2s derived from human PBMCs[1].
SM-17 (5 μg/mL) inhibits IL-25-driven ERK1/2 and STAT5 activation in human PBMCs[1].
SM-17 (1.25-5 μg/mL) dose-dependently restores filaggrin expression in human HaCaT keratinocytes cocultured with alarmin cocktail-stimulated human PBMCs, with the strongest effect at 5 μg/mL[1].

Result:Significantly reduced lung pathology scores compared to IgG4 control, with thinner bronchiole linings and fewer infiltrated cells.
Significantly suppressed goblet cell, mast cell, and eosinophil counts in lung tissue, with effects comparable to dexamethasone on goblet cell hyperplasia and eosinophil infiltration.
Strongly inhibited lung collagen deposition (a measure of airway remodeling), an effect not seen with dexamethasone.
Effectively abrogated HDM-induced IL-4, IL-5, and IL-13 levels in BALF, with inhibitory effects comparable to dexamethasone.
Strongly downregulated the number of type 2 innate lymphoid cells (ILC2) in BALF.
Identified 5 mg/kg as the optimal dose.

Flow cytometric analysis of 1X106 HepG2 cells labeling IL-17RB with SM-17 (HY-P991266, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/200 for an hour at 4℃. AF488-conjugated Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG4 kappa Isotype Control (HY-P99003, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
