2,2-Diphenyltetrahydrofuran (DPTHF) acts as a TRPV3 channel antagonist and is a structural analog of 2-Aminoethyl diphenylborinate (HY-W009724). 2,2-Diphenyltetrahydrofuran shows weak inhibitory activity against TRPV1 and TRPV2. It reduces hedonic feeding in mice exposed to Olanzapine (HY-14541). 2,2-Diphenyltetrahydrofuran can be used in the research of nervous system-related diseases.

2,2-Diphenyltetrahydrofuran (DPTHF) exerts multiple regulatory effects in HEK293 cells overexpressing TRPV3: it reversibly blocks TRPV3 calcium influx and whole-cell currents at ± 80 mV induced by 2-Aminoethyl diphenylborinate (HY-W009724) and DPBA; meanwhile, it directly and reversibly blocks heat-induced TRPV3 I2-phase currents; it also inhibits store-operated calcium entry in mammalian cells[2].
2,2-Diphenyltetrahydrofuran (100-500 μM) only weakly inhibits TRPV1 and TRPV2[2].

Animal Model:BALB/c (female, 18-23 g, metabolic alteration induced by 6 mg/kg olanzapine administered p.o. daily for 6 weeks)[1]

Downregulated TRPV3 mRNA in NAc, hypothalamus and VTA at both 10 mg/kg and 20 mg/kg, without altering TRPV1 mRNA in these regions.
Exerted no meaningful changes on body weight gain, weight change ratio, locomotor activity, body temperature, vWAT weight, adiposity index, serum glucose, insulin, HOMA-IR, OGTT-AUC, serum adiponectin, ghrelin, leptin, lipid profiles and serum IL-6 / NF-κB at two doses.
