HDAC6-IN-89
HDAC6-IN-89 is a selective HDAC6 inhibitor (IC50 values of 0.003, 0.031, 0.039, 0.046, 0.050, 0.057, and 3.30 μM for HDAC6, HDAC1, HDAC3, HDAC2, HDAC8, HDAC10, and GLS1, respectively). HDAC6-IN-89 increases Histone/Tubulin acetylation and blocks glutaminase-mediated conversion of glutamine to glutamate and glutamine metabolism. HDAC6-IN-89 induces ROS, Apoptosis, and cell cycle arrest. HDAC6-IN-89 exhibits anticancer activity against lung cancer, prostate cancer, and colon cancer.

In Vitro
HDAC6-IN-89 is a potent HDAC inhibitor with strongest potency against HDAC6 (IC50 0.003 µM) and selectivity over other HDAC isoforms[1].
HDAC6-IN-89 shows no antiproliferative activity against L02 cells with an IC50 > 100 µM[1].
HDAC6-IN-89 (0.2-5 µM; 24 h) increases α-tubulin and histone H3 acetylation in CT26 cells without altering total α-tubulin or histone H3 expression[1].
HDAC6-IN-89 (2 h) is relatively stable in human and mouse plasma, with 92.2% and 91.4% remaining after 2 h[1].
HDAC6-IN-89 has moderate rat liver microsomal stability with a half-life of 1.04 h and CLint of 22.11 µL/min/mg[1].
HDAC6-IN-89 shows no reversible inhibition against CYP enzymes[1].
HDAC6-IN-89 (10 µM) shows no obvious time-dependent inhibition of CYP1A2, CYP2A6, CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP2D6, or CYP2E1, with a measurable time-dependent inhibition tendency for CYP3A4[1].

Parmacokinetics
In Vivo
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
-
Animal Model:BALB/c nude (6-8 weeks old)[1]
-
Dosage:50 mg/kg
-
Administration:i.p.; once daily; 28 consecutive days
-
Result:At 50 mg/kg, a tumor growth inhibition rate of 91.10% was achieved based on final tumor weight.
Tumor volume and tumor weight were reduced relative to vehicle.
In tumor tissue, Ac-α-tubulin and Ac-Histone H3 levels were increased.
The proportions of tumor-infiltrating lymphocytes, CD4+ T cells, CD8+ T cells, activated CD8+ T cells, NK cells, proliferating NK cells, tumor-associated macrophages, and M1 macrophages were increased, while exhausted CD8+ T cells and M2 macrophages were decreased; these effects were most pronounced in the 50 mg/kg group.
