α-Rubromycin
α-Rubromycin (Collinomycin) is an antibiotic isolated from the mycelia of Streptomyces collinus. α-Rubromycin also acts as a DNA polymerase inhibitor, with an IC50 of 0.91 μM against bovine mammalian DNA polymerase. α-Rubromycin binds to the active region of DNA polymerase β, competes with nucleotide substrates for binding, and interferes with template-DNA interactions. α-Rubromycin inhibits the activities of telomerase, retroviral reverse transcriptase and terminal deoxynucleotidyl transferase, as well as the growth of Staphylococcus aureus. α-Rubromycin can be used in studies related to bacterial infections.

In Vitro
α-Rubromycin potently and selectively inhibits mammalian DNA polymerase α and β, with the strongest inhibitory effect on rat DNA polymerase β (IC50 = 0.45 μM). Its mechanism of action involves binding to the 31 kDa catalytic domain and competing with nucleotide substrates for binding, while it does not interfere with the binding of DNA templates[1].
α-Rubromycin (compound 1) inhibits M-MLV RT with an IC50 of 7.34 μM; it shows almost no activity against human immunodeficiency virus type 1 (HIV-1) reverse transcriptase, with an IC50 > 200 μM; it also exhibits extremely weak inhibitory effect on human telomerase, with an IC50 > 200 μM[2].
