Calamenene is a sesquiterpene compound. Calamenene promotes dendritic cell maturation, upregulates CD1a, CD80, CD83, CD86, HLA-DR and CCR7 on the cell surface, reduces endocytic activity, enhances T cell-stimulating capacity, drives Th1 polarization through the secretion of IL-12, induces IFN-γ production, decreases IL-4 generation, and triggers intracellular Ca2+ mobilization as well as dendritic cell migration towards MIP-3β. Calamenene exerts bacteriostatic and bactericidal growth-inhibitory effects against pathogenic *Vibrio harveyi*. Calamenene can be used in studies related to cancer and bacterial infections.

In Vitro

Calamenene (0.1-10 μM; 2 days) dose-dependently upregulates the expression of maturation markers (CD1a, CD80, CD83, CD86, HLA-DR) and reduces the endocytic activity in human monocyte-derived dendritic cells[1].
Calamenene (10 μM; 2 days) induces the maturation of human monocyte-derived dendritic cells. Following stimulation with CD40-L, these cells secrete high levels of Th1-promoting cytokines IL-12p70 and IL-6, as well as low levels of the anti-inflammatory cytokine IL-10, driving allogeneic naive CD4+ T cells to differentiate into Th1 cells that secrete high levels of IFN-γ and low levels of IL-4[1].
Calamenene (10 μM; 2 days) induces the maturation of human monocyte-derived dendritic cells, upregulates the expression of CCR7, enhances their migratory capacity toward MIP-3β, and exhibits stronger intracellular Ca2+ mobilization in response to MIP-3β[1].
Calamenene (0.1-5.0 µg/mL; 1 h at room temperature, followed by 24 h at 28 °C post-plating) inhibits the growth of Vibrio harveyi MMRF1107, and the bacterial mortality increases in a concentration-dependent manner[2].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Migration Assay [1]

Cell Line: human monocyte-derived dendritic cells
Concentration: 10 μM
Incubation Time: 2 days
Result: Enhanced migration to MIP-3β, and showed increased intracellular Ca2+ mobilization in response to MIP-3β, indicating a mature, migratory phenotype.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
