Lig4-4
Lig4-4 is an orally active and selective TREK-1 inhibitor, with an IC50 of 2.06 μM in rats. Lig4-4 improves neuronal viability, reduces cell apoptosis by upregulating cleaved-caspase-3 and downregulating Bcl-2, and decreases infarct volume. Lig4-4 exerts a neuroprotective effect against cerebral ischemia injury. Lig4-4 can be used in studies related to cerebral ischemia.

In Vitro
Lig4-4 potently and selectively inhibits the rat TREK-1 channel in CHO/TREK-1 cells, with an IC50 of 2.06 μM[1].
Lig4-4 (1-10 μM; 2 h OGD, 24 h reoxygenation) enhances the viability of in vitro cultured rat cortical neurons subjected to OGD/R injury and reduces the apoptosis level of in vitro cultured rat cortical neurons with OGD/R injury[1].
Lig4-4 (10 μM; 2 h OGD, 24 h reoxygenation) reverses the oxygen-glucose deprivation/reoxygenation (OGD/R)-induced upregulation of cleaved caspase-3 expression and downregulation of Bcl-2 expression in rat cortical neurons, without altering Bax expression[1].
