CRBN, or Cereblon protein, is the substrate recognition subunit of the Cullin 4-RING E3 ubiquitin ligase complex. Immunomodulatory drugs (thalidomide and its derivatives) directly bind to CRBN, altering its substrate specificity and enabling it to recognize and degrade transcription factors such as IKZF1/3, thereby exerting anti-tumor effects.
