Tonantzitlolone
Tonantzitlolone is a diterpenoid natural product found in Stillingia lineata ssp. lineata, and is a selective and reversible agonist of human TRPC1/4/5 ion channels. Tonantzitlolone reduces cancer cell viability and shows selectivity markedly restricted to a single cancer cell line. Tonantzitlolone can be used in research related to Merkel cell carcinoma and renal cell carcinoma.

In Vitro
Tonantzitlolone (TZL) (100 nM; 200-300 s) induces Ca2+ influx in A498 renal cell carcinoma cells that natively express TRPC1/4 channels; furthermore, the Ca2+ influx induced in TRPC5-expressing HEK 293 cells is blocked by 30 nM Pico145 (HY-101507)[3].
Tonantzitlolone (3-3000 nM) activates overexpressed TRPC4, TRPC5, TRPC4-TRPC1, and TRPC5-TRPC1 channels in HEK 293 cells, with EC50 values of 123.1, 83.6, 140.6, and 61.6 nM, respectively[3].
Tonantzitlolone activates TRPC5 channels in whole-cell recordings from HEK 293 cells, with an EC50 value of 76 nM at +100 mV and 64 nM at -100 mV[3].
Tonantzitlolone (1 µM) activates TRPC5 channels when applied to the extracellular surface of outside-out patches, but not when applied to the intracellular surface of inside-out patches[3].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
