ML 2-14 is a PROTAC targeting BRD4 with a C4 alkyl linker. ML 2-14 consists of the E3 ligase ligand EN219 (HY-115715) (bule part), the target protein ligand JQ-1 (HY-13030) (red part), and the PROTAC linker (balck part). ML 2-14 can effectively degrade BRD4 in 231MFP breast cancer cells, and this effect can be reversed by the proteasome inhibitor Bortezomib (HY-10227) and the E1 activase inhibitor TAK-243 (HY-100487). ML 2-14 can be used for breast cancer-related research.

BRD4 |
ML 2-14 (1 nM-1 μM; 8 h) significantly degraded BRD4 in 231MFP breast cancer cells, with DC50 values of 36 and 14 nMfor the long and short isoforms of BRD4, respectively[1].
The degradation of BRD4 by ML 2-14 (100 nM; 8 h) in 231MFP cells could be blocked by Bortezomib (4 μM; 30 min prior) and the E1 activating enzyme inhibitor TAK-243 (10 μM; 8 h prior)[1].

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
