Cancer Stem Cells
Cancer stem cells (CSCs) are populations of cancer cells that have the properties of stem cells i.e. self-renewal and ability to generate differentiated progenies. CSCs have high plasticity, which changes their phenotypic and functional appearance. They have important roles in tumor development, expansion, resistance, relapse and metastasis. Multiple studies have shown that CSCs originate from non-malignant stem or progenitor cells. Inhibition of developmental signaling pathways that are critical for stem and progenitor cell homeostasis and function has important implications in strategies to target CSCs in cancer research.
Studies have shown that CSCs develop several mechanisms to protect themselves from toxins and genotoxic stress, including enhanced DNA damage repair capacity, increased expression of drug transporters, maintenance of a low reactive oxygen species (ROS) environment, and recruitment of a protective niche. Therefore, targeting signaling pathways that are required to maintain stem cells is the current therapeutic strategy for CSCs. For example, hedgehog pathway, Notch and Wnt signaling pathways.
Newly developed drugs targeting surface markers of CSCs opens the new avenues for cancer therapy, such as CD44, CD133, EpCAM, Sox2, and Oct-3/4.

Gigantol exerts anti-lung cancer activity by inducing ferroptosis (Ferroptosis) via the SLC7A11-GPX4 axis. Gigantol restores the sensitivity of mcr-harboring multidrug-resistant bacteria to colistin. Gigantol ameliorates carbon tetrachloride-induced liver injury by inhibiting the activation of the JNK/cPLA2/12-LOX inflammatory pathway. Gigantol promotes cholesterol metabolism and progesterone biosynthesis in Leydig cells. Gigantol can be used in studies related to diseases such as lung cancer, multidrug-resistant Gram-negative bacterial infections, and acute liver injury.

STAT3-IN-11
0 ImagesSTAT3-IN-11 (7a) is a selective STAT3 inhibitor that inhibits the phosphorylation of STAT3 at site pTyr705. STAT3-IN-11 inhibits the phosphorylation of downstream genes (Survivin and Mcl-1) without affecting its upstream tyrosine kinases (Src and JAK2) levels and p-STAT1 expression. STAT3-IN-11 can induce cancer cell apoptosis, which is potential for the discovery of effective STAT3 inhibitors and antitumor agents against cancers.

BTX161
0 ImagesBTX161, a Thalidomide (HY-14658) analog, is a molecular glue degrader targeting CK1α. BTX161 recruits CK1α and the CK1α-FAM83 complex to the Cul4ACRBN E3 ligase complex, thereby driving ubiquitination and proteasomal degradation. BTX161 reduces FAM83G abundance through CK1α-FAM83G co-stability, slightly decreases FAM83H levels, but has no effect on the expression of FAM83B. BTX161 activates DNA damage response (DDR) and upregulates Wnt target genes including MYC. BTX161 can be used in research related to colorectal cancer, type b myelomonocytic leukemia and acute myeloid leukemia.
