DpC is an orally active iron chelator and anticancer agent. DpC induces lysosomal membrane permeabilization, redox-active iron/copper complex formation, oxidative stress, and Apoptosis through carrier/receptor-mediated uptake and Pgp-mediated lysosomal transport, while upregulating NDRG1 to inhibit oncogenic signaling and tumor progression, and modulates pathways such as JNK, IκBα/NF-κB, c-Src, and EGFR/HER2/HER3. DpC selectively inhibits tumor cell proliferation and can be used in research on neuroblastoma, kidney disease, prostate cancer, and pancreatic cancer.

DpC (serial dilution; 24-72 h) inhibits the proliferation of PANC-1, PC3, DMS-53, DU-145, and MDA-MB-231 cells, with IC₅₀ values at 72 h of 0.096, 0.007, 0.020, 0.007, and 0.008 μM, respectively^[1].
DpC (0.125-8 × IC₅₀; 72 h) combined with standard drugs shows synergistic effects at a lower frequency than sequential exposure in PANC-1, PC3, DMS-53, DU-145, and MDA-MB-231 cells, and DpC combined with Dp44mT (HY-18973) exhibits antagonism in all five cell lines, with DU-145 cells showing the highest CI of 16.009^[1].
DpC (25 μM; 2 h) uptake by SK-N-MC cells is temperature- and energy-dependent and is competitively inhibited by excess Dp44mT^[1].
DpC (2.5-250 μM; 24 h) is highly active against SK-N-LP neuroblastoma cells (IC₅₀ < 2.5 μM), whereas it shows much higher IC₅₀ values against MSC (145.23 μM), MIHA (165.73 μM), HK2 (17.27 μM), and H9C2 (> 250 μM) after 24 h^[2].
DpC (25 μM; 12-24 h) upregulates Cygb and Ngb in HK2 and SK-N-LP cells at 24 h, but slightly decreases their expression in MSC and H9C2 cells^[2].
DpC (5 μM; 24 h) induces NDRG1 and inhibits c-Src activation at Tyr416 in DU145 cells without altering total c-Src^[4].
DpC (10 µM) upregulates NDRG1 and reduces EGFR protein levels and membrane localization in PANC-1 and HT-29 cells^[5].

DpC (25 μM; 24 h) induces apoptosis in all four neuroblastoma cell lines, with SK-N-AS showing the highest proportion of early apoptosis (42.3%) and SK-N-LP showing the highest proportion of late apoptosis (27.4%)^[2].
DpC (25 μM; 24 h) increases Ngb, Cygb, cleaved caspase 3/9 and p-JNK/total JNK in SK-N-LP cells after 24 h, while decreasing IκBα^[2].
DpC (25 μM; 24 h) cytotoxicity in SK-N-LP cells is partially dependent on p38, JNK, NF-κB, and caspase activity, but not on ERK^[2].

Others

DpC does not show obvious cytotoxicity against HK-2 human proximal tubular epithelial cells and L929 cells^[3].

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Protocol 1

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Solubility: ≥ 2.08 mg/mL (5.88 mM); Clear solution

This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
