Cancer Immunotherapy
Cancer immunotherapy (CIT) is a type of biological therapy, aiming to improve anti-tumor immune responses with fewer off-target effects than chemotherapy. Several types of immunotherapy include: oncolytic virus therapies, cancer vaccines, cytokine therapies, adoptive cell transfer (ACT), and immune checkpoint inhibitors (ICIs). In particular, ICIs and ACT have obtained immense clinical response, but their efficacy varies from person to person. Immune cells can be harnessed to eliminate tumor cells, such as T cells, B cells, NK cells, and myeloid cells. T cells have potent tumor-killing capability, therefore, a plethora of cancer immunotherapy research have focused on inducing T-cell-mediated anti-tumor responses. CTLA-4 and PD-1 are found on the cell surface of T cells as co-inhibitory receptors. The breakthrough in cancer immunotherapy results from the identification and subsequent targeting of checkpoint mechanisms in T cells with monoclonal antibodies against CTLA-4 and programmed death-ligand 1/programmed death-1 (PD-L1/PD-1).
Several types of cancers (e.g., melanoma, mismatch repair-deficient cancers, bladder cancer, and non-small cell lung cancer) have achieved significant clinical responses in T-cell checkpoint blockade therapies. However, single-mode immunotherapy faces challenges such as low immune response, low tumor infiltration, and complex immunosuppressive tumor microenvironment. Recently, combined therapies based on tumor immunity have received extensive attention in the research of enhancing tumor cells immunogenicity and inhibiting their growth. For example, anti CTLA-4 and PD-1, immune checkpoint blockade (ICB) combined with chemotherapy, anti-angiogenic drugs and kinase inhibitors.

PF-04217903
0 ImagesPF-04217903 is an orally active, highly selective ATP-competitive c-Met kinase inhibitor with a Ki value of 4.8 nM and a Kd value of 4.5 nM. PF-04217903 blocks c-Met and HGF signaling pathways, inhibits MET phosphorylation, and blocks downstream MAPK, PI3K/AKT and PLCγ1 pathways. PF-04217903 suppresses tumor proliferation, survival, migration, invasion, angiogenesis and metastasis, induces apoptosis, and enhances efferocytosis, Annexin A1 expression and resolution of inflammation. PF-04217903 retains activity against several c-Met mutants (M1131T, V1220I, H1094R). PF-04217903 increases the incidence of subarachnoid hemorrhage and reduces survival rate without altering aneurysm formation, and also prevents lymph node metastasis induced by VEGF inhibition. PF-04217903 is applicable to research related to tumors (pancreas, stomach, lung, brain, colon, breast, kidney, melanoma, etc.), intracranial aneurysms and inflammatory diseases (gouty arthritis, neutrophilic pleuritis).

Tropisetron
0 ImagesSynonyms: SDZ-ICS-930 free baseTropisetron is an orally active 5-HT3R antagonist (Ki = 5.3 nM) as well as being a potent and selective α7 nicotinic partial agonist (EC50 = 1.3 μM). Tropisetron prevents phosphorylation and activation of the p38 MAPK. Tropisetron inhibits both IL-2 gene transcription and IL-2 synthesis in stimulated T cells. Tropisetron inhibits the binding to DNA and the transcriptional activity of NFAT and AP-1. Tropisetron is anti-inflammatory and antiemetic. Tropisetron has antitumor and neuroprotective effects. Tropisetron can be studied in research for diseases including hemorrhagic cystitis, chronic joint inflammation, lung cancer and chronic cerebral hypoperfusion.
