OptoDArG
OptoDArG is a photoswitchable diacylglycerol analog that serves as an optical lipid clamp probe. OptoDArG undergoes rapid trans-cis photoisomerization under UV/blue light, with the trans form being inactive, and can be delivered directly to the cell membrane without genetic modification. Cis-OptoDArG regulates the activity of channels such as TRPC3, TRPC6, and TRPL by altering the properties of the cell membrane lipid bilayer, thereby triggering sodium channel-dependent action potentials, PKCα translocation, mechanically sensitive currents, and various other downstream electrophysiological and cellular effects. OptoDArG has activity in identifying lipid-gated pores by controlling TRPC3 channels, and can serve as a tool for studying the lipid-sensing mechanisms of TRPC channels.

Biological Activity
Description
In Vitro
OptoDArG (20 m%; 50 ms) UV-driven trans-to-cis photoisomerization increases bilayer capacitance by +4.54 pF and generates depolarizing optocapacitive currents, whereas blue-driven cis-to-trans isomerization decreases capacitance by −3.57 pF and generates oppositely directed currents in PLBs[1].
OptoDArG (60 µM; 1 h) in HEK293 NaV1.3 cells, UV light generates optocapacitive depolarization that triggers APs with 33.3% success, while blue light triggers Gd3+-sensitive mechanosensitive depolarizing currents and APs[1].
OptoDArG (30 μM; 30 s) is a light-controlled DAG photoswitch whose cis isomer rapidly and reversibly activates TRPC3 conductance in HEK293 cells expressing TRPC3[2].
OptoDArG (30 μM) facilitates photoactivation with a lower light-intensity threshold and reduced activation sigmoidicity (mean n = 2.9 at maximum light intensity) in HEK293 cells expressing TRPC3-G652A[2].
OptoDArG photoactivation sensitivity is reduced in HEK293 cells expressing TRPC6-G709A[2].
OptoDArG is a highly efficient photochromic ligand that facilitates activation of the TRPC3 G652A mutant in HEK293 cells via a lipid-sensing site near the selectivity filter using 365/430 nm light[4].
