VVD-849 is a RAS ligand. VVD-849 covalently binds to Cys242 in the RAS-binding domain of PI3K p110α and promotes RAS/PI3K interaction. VVD-849 partially inhibits pAKT (S473) in HER2-overexpressing tumors. VVD-849 can be used for the research of cancers such as breast cancer.

VVD-849 (2 µM; 2 h) exhibits excellent selectivity for cysteine residues in live Jurkat cells[1].
VVD-849 (4 h) enhances the interaction between KRASG12C and p110α in HEK293T cell lysates (cotransfected with p85α), with an IC50 of 0.02 µM[1].
VVD-849 (30 min) increases the level of pAKTS473 in H358 cells, with an IC50 of 0.04 µM[1].
VVD-849 (2 h) partially inhibits the level of pAKT (S473) in N87 (HER2-overexpressing) cells[1].
VVD-849 (1 h) binds to p110α C242 in Jurkat cell lysates, with a TE50 of 0.063 µM[1].
VVD-849 (1 h) shows binding to p110α C242 in Jurkat cell lysates as detected by pocket probe assay, with a TE50 of 0.1 µM[1].
VVD-849 partially inhibits AKT phosphorylation in HER2-overexpressing human gastric cancer N87 cells, which may result from its combined stimulatory effect on the RAS-p110α interaction and inhibitory effect on the HER2-mediated PI3Kα signaling pathway[2].
