Description

Obafluorin is a β-lactone antibiotic. Obafluorin inhibits threonyl-tRNA synthetase (PfThrRS) with an IC50 of 4.3 nM. Obafluorin inhibits the aminoacylation activity of EcThrRS and ObaO. Obafluorin exhibits Fe3+-enhanced antibacterial activity. Obafluorin can be used in studies related to infections caused by bacterial and parasites such as Plasmodium[1][2].

In Vitro

Obafluorin (Compound 1) (16-18 h) inhibits the growth of B. subtilis (MIC 4 μg/mL), MRSA (MIC 2 μg/mL), E. coli ATCC 25922 (MIC 256 μg/mL) and E. coli NR698 (MIC 4 μg/mL) in the spot lawn antimicrobial assay[1].
Obafluorin (0-5000 nM; 10 min preincubation, up to 10 min reaction) inhibits the aminoacylation activity of purified EcThrRS and ObaO, with IC50 values of 92 nM and 50 nM, respectively[1].
Obafluorin (0-5000 nM; 10 min preincubation; 10 min assay incubation) potently inhibits EcThrRS and PfThrRS in vitro, with IC50 values of 35 nM and 4.3 nM, respectively[2].
The antibacterial activity of Obafluorin (16-18 h; 2 mM Fe3+ added to agar) is significantly enhanced in the presence of Fe3+: the MIC against E. coli ATCC 25922 decreases by ≥256-fold, the MIC against P. aeruginosa PA01 decreases by ≥128-fold, and the MIC against MRSA decreases by 8-fold[2].
