In Vitro

PP30 is a selective mTOR active-site inhibitor with an in vitro IC₅₀ value of 80 nM, and it exhibits only extremely weak activity against other members of the PI3K family as well as some protein kinases including PKCα and PKCβ^[1].
PP30 (0.04-10 μM; pre-incubated prior to insulin stimulation) inhibits insulin-induced phosphorylation of Akt at the S⁴⁷³ and T³⁰⁸ sites in L6 myotubes, with a stronger inhibitory potency against phosphorylation at the S⁴⁷³ site^[1].
