PROTAC K-Ras Degrader-5 is a cereblon-based K-Ras PROTAC degrader with a DC50 of <100 nM for KRASG12D. PROTAC K-Ras Degrader-5 recruits KRASG12D to the cereblon E3 ubiquitin ligase complex for ubiquitination and subsequent proteasomal degradation. PROTAC K-Ras Degrader-5 (compound 69) (1-3 mg/kg; i.v.; twice weekly; once weekly; once every two weeks) effectively inhibits tumor growth in GP2d colorectal cancer mice, with all tumor inhibition rates exceeding 90%[1].
PROTAC K-Ras Degrader-5 AsPC-1 (0.3-3 mg/kg; i.v.; twice weekly; once weekly; once every two weeks) effectively inhibits tumor growth in AsPC-1, SW1990, HPAC and Panc04.03 pancreatic cancer xenograft mouse models[1].
