(E)-Methyl 4-coumarate (Methyl 4-hydroxycinnamate) is a phenolic compound and derivative of Cinnamic acid (HY-N0610A). (E)-Methyl 4-coumarate can be found in several plants, such as the leaves of Allium cepa and Morinda citrifolia L. (E)-Methyl 4-coumarate, when combined with Carnosic acid (HY-N0644), induces Apoptosis. (E)-Methyl 4-coumarate inhibits GSK3β activity and modulates inflammatory cytokine levels (increasing IL-10 and decreasing IL-4). (E)-Methyl 4-coumarate combined with Carnosic acid exhibits anticancer effects against acute myeloid leukemia. (E)-Methyl 4-coumarate ameliorates Plasmodium berghei NK65 infection.

(E)-Methyl 4-coumarate (5 µM; 72 h) synergizes strongly with Carnosic acid (HY-N0644) to reduce the number of viable cells in HL60, KG-1a, and U937 acute myeloid leukemia (AML) cell lines^[1].

In Vivo

(E)-Methyl 4-coumarate (10-60 mg/kg; i.p.; consecutive administration; 4 days) inhibits parasitaemia development in mice infected with Plasmodium berghei NK65, prolongs the median survival time of mice, modulates serum inflammatory cytokine levels (reduces TNF-α and IFN-γ, increases IL-10), and increases the level of pGSK3β (Ser9) in mouse liver to inhibit GSK3β activity^[2].

Animal Model:

Animal Model: ICR mice infected with Plasmodium berghei NK65^[2]
Dosage:

10 mg/kg, 30 mg/kg, 60 mg/kg
Administration:

Intraperitoneal injection (i.p), consecutive administration, 4-day cycle
Result:

Resulted in average parasitaemia suppression rates of 62.45 % (10 mg/kg), 68.44% (30 mg/kg), and 51.84% (60 mg/kg) respectively on day 4 post-infection.
Showed median survival times of 15 days, 19 days, and 17 days at doses of 10 mg/kg, 30 mg/kg, and 60 mg/kg, respectively.
Reduced serum pro-inflammatory cytokines TNF-α and IFN-γ to 0.1-fold and 0.4-fold of the control group respectively, increased anti-inflammatory cytokine IL-10 to 3.0-fold of the control group, and decreased IL-4 to 0.6-fold of the control group.
Increased the level of pGSK3β (Ser9) in the liver of infected mice by 7.0-fold compared to the control group, indicating inhibition of GSK3β activity.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

Protocol 1

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

Solubility: ≥ 2.5 mg/mL (14.03 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.

Protocol 2

Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)

Solubility: ≥ 2.5 mg/mL (14.03 mM); Clear solution

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).

Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
