HaloPROTAC-E is a SGK3/VPS34 HaloPROTAC degrader that degrades Halo-tagged SGK3 and VPS34, with a DC50 of 3-10 nM in HEK293 cells. HaloPROTAC-E induces ubiquitination and proteasomal degradation of target proteins. HaloPROTAC-E blocks downstream phosphorylation of SGK3 substrate NDRG1. HaloPROTAC-E is applicable for cancer research.

HaloPROTAC-E (1-1000 nM; 48 h) potently induces dose-dependent degradation of endogenous SGK3-Halo and Halo-VPS34 in HEK293 cells, with a DC50 of 3-10 nM[1].
HaloPROTAC-E (300 nM; 10-240 min) induces rapid, reversible degradation of endogenous SGK3-Halo (50% degradation within 20-30 min) and VPS34-Halo (50% degradation within 1-2 h) in HEK293 cells[1].
Degradation of endogenous SGK3-Halo and Halo-VPS34 in HEK293 cells induced by HaloPROTAC-E (300 nM; 6 h) depends on the Cullin2-VHL E3 ligase pathway and proteasomal activity[1].
HaloPROTAC-E (300 nM; 0-24 h) mediates the degradation of endogenous SGK3-Halo and Halo-VPS34 in HEK293 cells, thereby functionally blocking the downstream SGK3 signaling pathway and reducing the phosphorylation level of NDRG1 Thr346[1].

This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
