BTX-7312 is a potent SOS1 molecular glue degrader. BTX-7312 forms a ternary complex by linking the target protein SOS1 with the E3 ubiquitin ligase cereblon, inducing ubiquitination and degradation of SOS1, thereby blocking the RAS/MAPK and PI3K/AKT signaling pathways and consequently inhibiting cell proliferation. BTX-7312 can be used for research on KRAS-driven cancers.

In Vitro
BTX-7312 synergizes with Sotorasib (HY-114277), Trametinib (HY-10999), Afatinib (HY-10261), and MRTX1133 (HY-134813) in 3D proliferation assays across KRAS-mutant cell lines[1].
BTX-7312 (0.3 nM-2 μM; 24 h) dose-dependently degrades SOS1 protein in MIA PaCa-2 and LoVo cells, with DC50 values of 0.5 nM and 3.9 nM, respectively, and does not affect the expression of proteins such as SOS2[1].
BTX-7312 (50-200 nM; 1-6 h) induces time-dependent degradation of SOS1 protein in MIA PaCa-2 and LoVo cells[1].
BTX-7312 (50-200 nM; 6 h) degrades wild-type SOS1 in H358 parental cells and in cells with knockout and knock-in of SOS1 mutants, but does not degrade H905V or N879A/H905V mutant SOS1[1].
BTX-7312 (0.8-500 nM; 6-24 h) reduces the levels of downstream signaling proteins pERK and pS6 (S235/S236) in MIA PaCa-2, LoVo, and A549 cells[1].
BTX-7312 has a slight inhibitory effect on cell viability in HEK293 and NHLF cells (IC50 > 5 µM)[1].
BTX-7312 (0.3 nM-2 μM; 24 h) does not induce the degradation of proteins such as EGFR, SHP2, GRB2, and KRAS in MIA PaCa-2 cells[1].
BTX-7312 (0.2-5 µM; 24 h) does not affect CK1α protein expression in MIA PaCa-2 and LoVo cells[1].

Select the appropriate dissolution method based on your experimental animal and administration route.
- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (2.70 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (2.70 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
