IKKβ-IN-2

IKKβ-IN-2 is an orally effective IκB kinase β (IKKβ) inhibitor. IKKβ-IN-2 inhibits the activity of IKKβ, thereby blocking TNFα-induced downstream NF-κB pathway activation and transcription, thus exerting inhibitory effects on inflammation and tumor growth. IKKβ-IN-2 can be used for research on colon cancer and pancreatic cancer.

IKKβ-IN-2 (compound 39-100) inhibits the growth of HCT116, SW620, HT29, DLD1, MiaPaCa2, AsPC1, Suit2, and S2013 cells with GI₅₀ values of 3.1-8.4 µM^[1].

Apoptosis

IKKβ-IN-2 (5-20 μM) dose-dependently decreased the phosphorylation level of IκBα (p-IκBα), downregulated the anti-apoptotic protein Mcl-1, and induced PARP cleavage in cells in vitro^[1].

Others

IKKβ-IN-2 inhibits TNFα-induced NFκB transcriptional activity in cell-based experiments with an inhibition rate of 83.8%^[1].

In Vivo

Anti-cancer

IKKβ-IN-2 (compound 39-100) (40 mg/kg; 2 weeks) significantly inhibited colon cancer tumor growth in the HCT116 colon cancer subcutaneous xenograft mouse model^[1].
IKKβ-IN-2 (40 mg/kg; p.o.; once daily; 4 weeks) reduced tumor volume and decreased tumor mass in a syngeneic orthotopic pancreatic cancer mouse model^[1].
