FzM1.8 is a Wnt/β-catenin signaling pathway agonist and an allosteric agonist of Frizzled 4 (FZD4), with a pEC50 of 6.4 for FZD4. FzM1.8 stabilizes a conformation with increased affinity for heterotrimeric G proteins, promoting Gβγ release and subsequent PI3K activation. FzM1.8 activates the AKT pathway downstream of FZD4. FzM1.8 does not affect FZD4 maturation or plasma membrane localization and does not induce FZD4 endocytosis. FzM1.8 upregulates the WNT-responsive genes C-myc and Cyclin D1, while leaving lgr5 expression unchanged. FzM1.8 can be used for colon cancer research.

FzM1.8 (2 µM) counteracts 48 h SMG-induced Wnt/β-catenin pathway inhibition and up-regulates P-gp expression in Caco-2 cells[1].
FzM1.8 activates a FZD4/PI3K signaling axis involving heterotrimeric G proteins and CBP/p300 in HEK293 cells, as its activity is blocked by Ly294002, I-CBP 112, Suramin, and Gallein but not by PKC or NFAT inhibitors[2].
Expression of a dominant-negative AKT mutant (K179M) abolishes FzM1.8-induced WRE activation in HEK293 cells, confirming AKT's essential role in the pathway[2].
The FZD4-wt/FZD4-T425D hetero-oligomer mimics the FzM1.8-bound state of FZD4, as it biases signaling toward the PI3K pathway, but the FZD4-T425D homocomplex does not[2].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.08 mg/mL (6.45 mM); Clear solution
This protocol yields a clear solution of ≥ 2.08 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.8 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
