ODN 2007
Based on 1 Customer Validation
ODN 2007 is an immunostimulatory CpG oligodeoxynucleotide. ODN 2007 activates TLR21 (the chicken homolog of TLR9), triggering innate signaling. ODN 2007 induces CpG motif-specific, dose-dependent 2'5'-A synthetase. ODN 2007 stimulates lymph node IFN-α/IFN-γ secretion. ODN 2007 binds Hsp90α, activates ERK2/AKT, and induces Th1 cytokine/chemokine mRNA (IL-6, MIP-3α) and NO. ODN 2007 can be used in research on Escherichia coli infection.

In Vitro
ODN 2007 (200 ng/mL; 16 h) induces chBMDC maturation, whereas ODN 2007-loaded CpG-NP treated at 200 ng/mL for 16 h produces a higher proportion of DC-like cells and higher CD40/CD80/CCR7 expression[1].
ODN 2007 (200 fmole; 20 min) directly binds to HD11 cytoplasmic and membrane proteins, with a stronger complex formed in the cytoplasmic F1 fraction[4].
ODN 2007 (10 µg/mL; 0-12 h) induces ERK2 phosphorylation starting at 30 min and AKT phosphorylation starting at 3 h in HD11 cells, and Hsp90, ERK1/2, and PI3K inhibitors reduce these induced phosphorylations[4].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Line:HD11 avian macrophage cells
-
Concentration:10 µg/mL (ODN 2007 sodium); 2 µM/mL (geldanamycin); 50 µM/mL (PD98059); 10, 50, 100 µM/mL (LY294002)
-
Incubation Time:0, 1/2, 1, 3, 6, 12 h (stimulation); 60 min (inhibitor pre-incubation)
-
Result:No significant difference was observed in Hsp90α or Hsp90β expression in stimulated or control HD11 cells.
A significant increase in phosphorylated ERK2 was detected in HD11 cells at 1/2 h, 1 h, 6 h, and 12 h.
A significant increase in phosphorylated AKT was observed at 3 h, 6 h, and 12 h; AKT phosphorylation started as early as 3 h, whereas ERK2 phosphorylation increased as early as 30 min.
Pre-incubation with geldanamycin (2 µM/mL) reduced phosphorylated ERK2 and phosphorylated AKT compared to control.
Pre-incubation with PD98059 (50 µM/mL) reduced phosphorylated ERK2 and phosphorylated AKT.
Pre-incubation with LY294002 (50 µM/mL) reduced phosphorylated ERK2 and phosphorylated AKT.
In Vivo
ODN 2007 (4 μg; subcutaneous injection, interscapular back; primary immunization and two boosts; at 2, 5, and 9 weeks of age) loaded in CpG-NP-adjuvanted rRBD immunization increases anti-rRBD antibody titers and reduces IBV re-isolation in the lungs and kidneys of SPF chickens[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
-
Animal Model:specific pathogen-free (SPF) (age at prime 2 weeks, boosted at 5 and 9 weeks)[1]
-
Dosage:4 μg per chicken
-
Administration:s.c. at the back between wings; prime and two boosts; at 2, 5, and 9 weeks of age
-
Result:After the first boost, anti-rRBD antibody titer barely differed from the PBS control.
After IBV challenge, virus re-isolation rates were lung 4/5 and kidney 4/5.
-
Animal Model:specific pathogen-free (SPF) (age at prime 2 weeks, boosted at 5 and 9 weeks)[1]
-
Dosage:4 μg CpG per chicken
-
Administration:s.c. at the back between wings; prime and two boosts; at 2, 5, and 9 weeks of age
-
Result:After the first boost, anti-rRBD antibody titer was 5711, nearly 4-fold higher than controls.
After IBV challenge, virus re-isolation rates were lung 1/5 and kidney 2/5.
-
Animal Model:SuVolk (either sex; 3-4 months of age; average body weight 46.1 kg)[2]
-
Dosage:100 µg/kg (30% emulsigen/PBS); 100 µg/kg (PBS)
-
Administration:s.c.; repeated 6 days later
-
Result:Serum 2′5′-A synthetase levels increased after both primary and secondary injections.
Peak levels were consistently observed 4 days after each injection.
Serum 2′5′-A synthetase levels were consistently higher in sheep receiving emulsigen formulation than in sheep receiving PBS formulation.
-
Animal Model:SuVolk (either sex; age range 1 week to 4 months)[2]
-
Dosage:100 µg/kg (30% emulsigen); 100 µg/kg (PBS)
-
Administration:s.c.; single injection
-
Result:At 48 h after injection, marked perivascular mononuclear cell infiltration was observed; formulation in emulsigen resulted in greater accumulation of mononuclear cells and eosinophils.
The majority of accumulating cells were CD172a+ myeloid cells and MHC class II+ cells; few T cells and few CD1+ cells were detected.
Injection sites stained intensely for IFN-α whether formulated in emulsigen or PBS.
Intense IFN-γ staining was observed only with emulsigen formulation, whereas PBS formulation showed faint IFN-γ staining.
