TL13-112 is a ALK PROTAC degrader. TL13-112 degrades the NPM-ALK protein by recruiting the CRBN E3 ligase, thereby blocking oncogenic signal transduction such as STAT3, ERK and AKT, and relieving the transcriptional repression of CD45, CCR7 and PI3Kδ. TL13-112 can be used in related research on anaplastic large cell lymphoma.

In Vitro
TL13-112 (100 nM; 12 h) induces potent degradation of ALK protein in TS and JB6 cells and specifically reduces the interaction between NPM-ALK and PTPN1/PTPN2[1].
TL13-112 (10-50 nM; 8-48 h) significantly degrades ALK protein in a time-dependent manner in TS, SU-DHL1 and JB6 cells, inhibits the level of downstream pALK (Y1604), and upregulates the cell surface expression and mRNA transcription levels of CD45 and CD45RO[2].
TL13-112 (50-100 nM; 8-72 h) degrades ALK protein in KARPAS-299, DEL, COST, and SU-DHL1 cells, upregulates the expression of PI3Kδ (p110δ) protein, and significantly upregulates the cell surface expression of CCR7 mRNA and protein, as well as the cell surface fluorescence intensity of CCR7[3].
