Y-27632 is a ROCK inhibitor with Ki values of 220 nM and 300 nM for ROCK1 and ROCK2, respectively. Y-27632 exerts anti-inflammatory and immunomodulatory effects in systemic lupus erythematosus models by inhibiting the ROCK/NF-κB pathway. Y-27632 enhances autophagy by inhibiting the AKT/mTOR pathway, thereby inducing apoptosis apoptosis in oral squamous cell carcinoma. Y-27632 induces the formation of tunneling nanotubes in ARPE-19 cells and significantly enhances mitochondrial transfer through these channels. Y-27632 promotes neurite outgrowth in PC12 cells by activating the Rac1/NOX1/ROS/AKT/PAK1 signaling cascade.

    Customer Validation
     On day 0, hPSCs were seeded onto new Matrigel- coated 6-well plates in StemMACS iPS-Brew XF medium + 10 μM Y-27632. On days 1 to 4, NC induction medium was added and changed daily. On days 5 and 6, NC induction medium + 1 μM retinoic acid was added daily. On day 7, cells were dissociated and split 1:6 in NC maintenance medium + 1 μM retinoic acid + 10 μM Y-27632. On days 8 and 9, NC maintenance medium + 1 μM retinoic acid was refreshed daily. On day 10, cells were dissociated and split. From days 11 to 21, SMC induction medium was refreshed daily. On days 22 to 28, cells were maintained in Medium 231+ Smooth Muscle Growth Supplement. Confocal fluorescence imaging and immunostaining confirms the differentiation of hESC-3R into CMs (coexpression of troponin-T with TNNT2-GFP), ECs (coexpression of VE-cadherin with CDH5-mOrange), and SMCs (coexpression of SM22α with TAGLN-CFP).
