Quinidine gluconic acid is an orally effective antiarrhythmic agent. Quinidine gluconic acid blocks the fast inward sodium current in a use-dependent manner, blocks IKr in a reverse use-dependent manner, and blocks the ultra-rapid delayed rectifier potassium current with an EC₅₀ of 6.2 μM, while also inhibiting CYP2D6 and P-gp. Quinidine gluconic acid induces PARP cleavage, Caspase-3 activation, and increases the proportion of sub-G1 phase apoptotic (Apoptotic) cells. Quinidine gluconic acid can be used in research on ischemia/reperfusion-induced cardiac injury, drug-resistant leukemia, convulsions, and uterine sarcoma.

In Vitro

Quinidine gluconic acid (10 μM; 24 h) enhances the cytotoxicity of Paclitaxel (HY-B0015) against P-glycoprotein (P-gp)-positive MES-SA/DX5 uterine sarcoma cells (reducing the IC₅₀ of Paclitaxel to 80.56 nM), but does not alter the cytotoxicity of Paclitaxel against P-gp-negative MES-SA uterine sarcoma cells^[1].
Quinidine gluconic acid (10 μM; 24 h) significantly increases the proportion of sub-G1 apoptotic cells in Paclitaxel-treated P-gp-positive MES-SA/DX5 uterine sarcoma cells and enhances Paclitaxel-induced apoptosis^[1].
Quinidine gluconic acid inhibits the in vitro metabolism of Debrisoquine in rat liver microsomes^[3].

Did not significantly alter the cytotoxicity of Paclitaxel in MES-SA cells, with Paclitaxel IC₅₀ remaining comparable to Paclitaxel alone (19 nM vs. 18 nM).
Strongly potentiated Paclitaxel-induced cytotoxicity in MES-SA/DX5 cells, reducing the IC₅₀ of Paclitaxel from 13851 nM to 81 nM.
Showed no significant cytotoxicity up to 100 μM in either cell line.\ Showed concentration-dependent cytotoxicity with docetaxel against MES-SA cells.

In Vivo

Quinidine (80 mg/kg; oral; single dose) gluconic acid potently inhibits amphetamine ring hydroxylation in male Lewis rats, reducing the excretion of p-hydroxyamphetamine to 7.2% and 24.1% of control levels at 24 h and 48 h, respectively, while increasing late-phase amphetamine excretion to 542% of control^[3].
Quinidine (10-30 mg/kg; intraperitoneal injection; single dose) gluconic acid significantly increases the pentylenetetrazol-induced tonic seizure threshold in male NMRI mice^[4].

Achieved total combined excretion of amphetamine and p-hydroxyamphetamine over 48 h equal to 79.7% of the administered dose, which was not significantly different from control groups.
