MAHS-4
MAHS-4 is an inhibitor of thymidylate synthase (TS) and dihydrofolate reductase (DHFR), with IC50 values of 1.918 μM and 83.53 μM, respectively. MAHS-4 inhibits thymidylate biosynthesis, reduces tetrahydrofolate pool maintenance, and interferes with extracellular matrix remodeling. MAHS-4 induces G2/M phase cell cycle arrest, activates endogenous caspase-dependent apoptosis, decreases MMP protein levels, inhibits cell migration, increases intracellular NO levels, and promotes autophagy activity. MAHS-4 is applicable for cancer-related research.

MAHS-3 induces cell cycle arrest and upregulates the expression of p21 and p27; it activates endogenous caspase-dependent apoptosis and regulates the levels of Bax, Cas-7, Cas-9, Bcl-2 and PARP1[1].
MAHS-4 exhibits dose-dependent antiproliferative activity in various cancer cell lines, shows cytostatic activity in some cancer cell lines, and has significantly higher cytotoxicity against breast cancer cells than normal cells and non-tumorigenic cells; moreover, it has better cancer cell selectivity than MAHS-3 (HY-187513)[1].
