Key Targets For Interfering With Viral Assembly, Release, And Maturation

Viral assembly, release, and maturation are late replication steps that convert viral genomes and structural proteins into infectious particles. In HIV-1, morphogenesis includes assembly, budding, and maturation, and the late phase includes Gag trafficking, genome encapsidation, Env incorporation, virion release, and protease-driven structural remodeling. These steps remain major antiviral targets because they expose vulnerable viral proteins and host cofactors after genome replication has already occurred[1][2].

HIV-1 Gag is the central assembly scaffold, and its domains coordinate plasma-membrane targeting, RNA packaging, particle formation, and recruitment of host ESCRT machinery. Tsg101 and the vacuolar protein sorting pathway are required for HIV-1 budding, and HIV-1 uses ESCRT proteins to complete membrane scission and release infectious particles. BST-2/tetherin blocks retrovirus release by retaining nascent virions at the cell surface, while viral antagonists such as Vpu counteract this restriction pathway[2][3][4].

Maturation provides another druggable checkpoint because HIV-1 protease cleaves Gag and Gag-Pol polyproteins into mature structural and enzymatic domains. Protease-mediated maturation converts immature particles into infectious virions, and inhibitors of protease or maturation disrupt this transition. Bevirimat stabilizes the immature Gag lattice and blocks late CA-SP1 processing, showing that maturation inhibitors can target substrate cleavage sites rather than only the protease active site[5][6].

Drug discovery now focuses on protease, capsid, Gag lattice, CA-SP1, ESCRT interfaces, tetherin antagonism, and virus-specific release enzymes. Influenza research highlights neuraminidase as a release target because neuraminidase structure and activity support virion release and functional balance with hemagglutinin. Remaining gaps include resistance, viral diversity, host-factor toxicity, and the need to inhibit conserved assembly or release mechanisms without disrupting essential cellular pathways[7][8].
