Cuproptosis: An Emerging Target In Cancer Therapy

Cuproptosis represents a distinctive regulated cell death mode elicited by mitochondrial copper. Specifically, this process entails the binding of copper ions to acylated proteins involved in the tricarboxylic acid (TCA) cycle within mitochondria. Such binding event triggers protein aggregation and the degradation of iron-sulfur cluster-containing proteins, ultimately resulting in cellular demise. Since its initial identification in 2022, cuproptosis has been closely associated with the field of materials science. Owing to its distinct activation pathway, it has also garnered substantial attention in the realm of cancer therapy [1] [2] [3] [4].

Tumor cells depend on aerobic glycolysis, which potentially enables them to escape cuproptosis. In contrast, targeting cuproptosis provides a novel strategy for overcoming tumor drug resistance. Additionally, inducing cuproptosis in tumor tissues can remodel the tumor microenvironment, thereby facilitating dendritic cell maturation and immune cell infiltration. Several cuproptosis inducers, including Elesclomol and Disulfiram, have demonstrated anticancer activities. Elesclomol selectively exerts cytotoxic effects on Cisplatin-resistant lung cancer cells by promoting reactive oxygen species (ROS) production. Disulfiram, on the other hand, markedly suppresses the proliferation of prostate cancer cells. These findings underscore the considerable potential of cuproptosis inducers for cancer treatment [5] [6].

At present, cuproptosis research remains in its early developmental phase. Notably, clinically available cuproptosis inducers such as Elesclomol and Disulfiram have not yielded satisfactory therapeutic outcomes in clinical trials. Therefore, future research and development endeavors ought to center on novel cuproptosis inducers or innovative induction strategies, encompassing natural products. Additionally, enhancing the delivery precision of drugs to tumor cells and improving the plasma stability of cuproptosis inducers should be prioritized in subsequent development efforts [6].
