Hidden Logic Behind STING-Driven Antitumor Immunity

STING-driven antitumor immunity describes cytosolic DNA sensing that converts tumor-derived DNA or treatment-induced DNA damage into type I interferon signaling, dendritic-cell activation, T-cell priming, and tumor immune infiltration. cGAS identifies cytosolic DNA and produces cGAMP, which activates STING and downstream type I interferon programs. Early cancer-immunity studies placed STING upstream of spontaneous antitumor T-cell responses and the T-cell-inflamed tumor microenvironment. Direct intratumoral STING activation induced regression of established tumors in mice and generated systemic immune responses, supporting STING agonism as an experimental strategy for cancer immunotherapy. Radiation studies further showed that STING, cGAS, dendritic-cell type I interferon induction, and adaptive immune responses form a linked pathway through which radiotherapy can promote antitumor immunity[1][2][3][4][5].
The main experimental logic is that STING signaling can bridge innate immune sensing and adaptive tumor control, but the response depends on cellular context, route of activation, and tumor state. Systemic non-nucleotide STING agonism with SR-717 activated STING and promoted antitumor immunity in preclinical models, while clinical trials of intratumoral MIW815/ADU-S100 showed pathway feasibility but limited antitumor responses, including minimal responses when combined with spartalizumab. Recent work refines the target-cell question by showing that conventional type 1 dendritic cells are essential for STING-mediated rejection of multiple established and metastatic murine tumors. The key unresolved gap is how to drive productive dendritic-cell licensing without chronic pathway rewiring, because chromosomal instability can chronically activate cGAS-STING and promote pro-metastatic signaling, and ENPP1 can degrade extracellular cGAMP to enable immune evasion. Future experiments should prioritize cell-selective STING delivery, dendritic-cell biomarkers, cGAMP metabolism, chromosomal-instability status, and combinations with radiation or checkpoint blockade[6][7][8][9][10][11].
