Stress Granules & Neurodegenerative Diseases

Stress granules (SGs) are dynamic membrane-less ribonucleoprotein assemblies that form in response to cellular stress, including oxidative stress, heat shock, viral infection, and metabolic perturbation. SGs transiently sequester untranslated messenger RNAs and associated RNA-binding proteins, thereby coordinating adaptive stress responses and translational control. With the emergence of liquid-liquid phase separation (LLPS) as a central organizing principle in cell biology, SGs have become a major focus of neurodegenerative disease research. Numerous proteins implicated in amyotrophic lateral sclerosis (ALS), frontotemporal dementia (FTD), Alzheimer’s disease, and related disorders localize to SGs and influence their assembly and dynamics, linking SG dysregulation to early pathogenic events in neurodegeneration[1][2][3][4][5].
Stress granule assembly is primarily driven by inhibition of translation initiation, with phosphorylation of eukaryotic initiation factor 2 alpha (eIF2α) during the integrated stress response representing a major regulatory pathway. Under stress conditions, untranslated mRNAs interact with RNA-binding proteins such as TIA1, G3BP1, TDP-43, and FUS to form SGs through multivalent molecular interactions. Physiological SGs are highly dynamic and normally disassemble after stress resolution. However, persistent stress, impaired proteostasis, and disease-associated mutations can promote SG stabilization and maturation into less dynamic assemblies. Mutations affecting TDP-43, FUS, hnRNPA1, and TIA1 alter SG dynamics and have been associated with pathological protein accumulation and neurotoxicity in ALS and FTD[2][3][4][6][7][8].
SG biology is widely investigated in studies of neurodegenerative disease mechanisms, therapeutic target discovery, and RNA metabolism. Current research directions include modulation of the integrated stress response, restoration of normal SG disassembly, and maintenance of ribonucleoprotein homeostasis. Because SGs are closely connected to phase separation, protein aggregation, and cellular stress signaling, they represent promising targets for mechanistic and translational research. Nevertheless, key questions remain regarding the long-term physiological functions of SGs in neurons, compositional heterogeneity among SG subtypes, and the causal relationship between SGs and pathological inclusions. Future studies combining live-cell imaging, single-cell approaches, and disease-relevant models are expected to improve understanding of SG regulation and support development of targeted interventions for neurodegenerative diseases[1][5][7][8][9].

Wright stain combines the acidic dye eosin (Eosin) with the basic dye methylene blue (Methylene Blue) to differentiate cellular structures. Giemsa stain consists of Azure II and eosin. Although the two stains have broadly similar staining mechanisms and color reactions, Giemsa stains the cytoplasm more strongly and shows its degree of basophilia more clearly. In blood and bone marrow specimens, it provides particularly effective staining of azurophilic, eosinophilic, and basophilic granules. However, its relatively intense nuclear staining can make fine intranuclear details difficult to distinguish. Because their staining properties are complementary, the two stains are often used together to offset their respective limitations.
