Migrasome: Cancer & Beyond Therapeutic Target

Migrasomes are a distinct class of extracellular vesicular structures generated during cell migration and were first described in 2015. Their formation depends on retraction fibers left behind migrating cells and on tetraspanin-enriched membrane domains. Migrasomes contain proteins, RNAs, lipids, and organelle-derived cargo and mediate intercellular communication through a process termed migracytosis. Since their discovery, migrasomes have emerged as an important signaling system alongside exosomes and microvesicles. Studies demonstrate roles in embryonic development, tissue homeostasis, immune regulation, organ morphogenesis, directional cell migration, and tissue repair, making migrasomes a rapidly expanding area of research in cell biology, cancer biology, and translational medicine[1][2][3][4].
Migrasome biogenesis is regulated by cell migration, integrin-mediated adhesion, and tetraspanins, particularly TSPAN4. TSPAN4-enriched membrane domains promote migrasome growth and stabilization, while cholesterol accumulation and membrane mechanical forces contribute to their formation. Migrasomes transport bioactive cargo including chemokines, signaling proteins, mRNAs, and mitochondria, influencing recipient-cell behavior after cargo release or uptake. During embryogenesis, migrasomes establish CXCL12 signaling gradients that guide cell migration. In mitochondrial quality-control pathways, damaged mitochondria can be selectively removed through migrasome-mediated disposal, a process termed mitocytosis. These findings position migrasomes at the intersection of cell migration, intercellular communication, and organelle homeostasis[1][2][5][6][7].
In disease biology, migrasomes are increasingly implicated in cancer progression, tumor microenvironment remodeling, inflammatory responses, cardiovascular disorders, and neurological diseases. Evidence indicates that tumor-derived migrasomes facilitate intercellular information transfer and influence tumor growth and metastatic behavior. Migrasomes are also being investigated as circulating biomarkers and therapeutic targets. However, major challenges remain, including the lack of standardized isolation methods, incomplete understanding of cargo-selection mechanisms, and limited knowledge of tissue-specific and disease-stage-dependent functions. Future studies integrating single-cell technologies, high-resolution intravital imaging, and multi-omics approaches will be required to define migrasome regulatory networks and evaluate their translational potential for disease diagnosis and therapeutic intervention[1][3][4][8][9].
